Cell Size and Palmitate - 1 - 14 C Turnover of Rabbit Marrow Fat
نویسنده
چکیده
Adipose tissue is a major component of normal rabbit marrow. Morphological considerations suggest an active role for this tissue in hematopoiesis. This hypothesis was tested by injecting 50 tCi of palmitate-1-14C intravenously into fed, hematologically normal New Zealand rabbits. The animals were sacrificed 24 hr later and the femoral marrow removed. Samples of subcutaneous and perinephric fat were taken for comparison. The fat cells were isolated by the Rodbell method and the diameters measured. Incorporation of the ‘4C-palmitate in the triglyceride fraction was determined and the composition of the fatty acids was measured by gas chromatography. The mean diameter of the marrow fat cell was 46 zm (mean cell volume 55 pI); the mean diameter of the perinephric fat cell 70 tm (mean cell volume 200 pI). ‘4C-Palmitate turnover per gram triglyceride was some fivefold greater in the marrow fat; however, when expressed on a cell basis, the turnover for the marrow and perinephric fat cell was similar. The marrow fat contained a higher concentration of unsaturated fatty acids. These findings suggest that there is greater lipolysis and lesser storage in the marrow fat than in the perinephric.

متن کاملBlastema from rabbit ear contains progenitor cells comparable to marrow derived mesenchymal stem cells
Rabbits have the capacity to regenerate holes in their ears by forming a blastema, a tissue that is made up of a group of undifferentiated cells. The purpose of the present study was to isolate and characterize blastema progenitor cells and compare them with marrow mesenchymal stem cells (MSCs). Five New Zealand white male rabbits were used in the present study. A 2-mm hole was created in the a...
متن کاملComparison of oxidative metabolism in starved, fat-fed and carbohydrate-fed rats.
1. Rats were starved for 48hr. or fed for 1 week on a high-fat or a high-carbohydrate diet. The effects of these dietary alterations on the rate of production of (14)CO(2) from trace amounts of [U-(14)C]glucose, [1-(14)C]palmitate or [1-(14)C]acetate administered intravenously were studied. 2. The oxidation of [(14)C]glucose was most rapid in the carbohydrate-fed condition and was decreased sig...
متن کاملMyocardial triglyceride turnover and contribution to energy substrate utilization in isolated working rat hearts.
The objective of this study was to determine the contribution of myocardial triglycerides to overall ATP production in isolated working rat hearts. Endogenous lipid pools were initially prelabeled (pulsed) by perfusing hearts for 60 min with Krebs-Henseleit buffer containing 1.2 mM [1-14C]palmitate. During a subsequent 60-min period (chase), hearts were perfused with either no fat, low fat (0.4...
متن کاملUptake and esterification of palmitrate by rat diaphragm in vitro.
The contribution of exogenously supplied palmitate to the intracellular palmitate pool and its role in esterification were studied for the intact rat diaphragm in vitro. Palmitate-1-(14)C attached to albumin in various molar ratios (nu) was taken up by the tissue in an initial rapid phase which led, after 4 min, to a steady-state level of tissue free fatty acid. The level was determined by nu b...
